چکیده

Mek inhibition and Erk knockout (KO) have quite distinct effects on pluripotency maintenance in mouse embryonic stem cells (ESCs). To test whether there is an Erk-independent function of Mek, RNA-sequencing (RNA-seq) is carried out on six samples, WT KH2 ESCs treated with or without PD0325901 (PD) for 48 h (KH2_PD and KH2, respectively), iErk1; Erk KO ESCs cultured in the presence of Dox (P0), 48 and 96 h after Dox withdrawal (P1 and P2, respectively), and iErk1; Erk KO ESCs cultured without Dox for 96 h, and treated with PD in the last 48 h (P2_PD). These RNA-seq data demonstrate that Mek inhibition has quite different effect on the transcriptional profile of mouse ESCs, compared to Erk KO. Moreover, a significant fraction of genes is regulated by Mek inhibition, regardless of the presence or absence of Erk, indicating an Erk-independent function of Mek. RNA-seq data are deposited in Gene Expression Omnibus (GEO) datasets under accession number GSE70304.

P-111: EGFR, ERK, MEK Genes Expression Level in Cumulus Cells of PCOS Women Compared with Healthy Women

Background Poly cystic ovarian syndrome (PCOS) is known as a common endocrine disorder in women at reproductive ages and may cause developmental abnormality in oocyte. ERK has found as a regulator protein of Gap junctions (GJ) function and the level of exchanges between two neighbors cells, for example oocyte and surrounding cumulus cells (CCs) in the mammalian ovary. Such exchange is essential...

Maintenance of Self-Renewal and Pluripotency in J1 Mouse Embryonic Stem Cells through Regulating Transcription Factor and MicroRNA Expression Induced by PD0325901

Embryonic stem cells (ESCs) have the ability to grow indefinitely and retain their pluripotency in culture, and this self-renewal capacity is governed by several crucial molecular pathways controlled by specific regulatory genes and epigenetic modifications. It is reported that multiple epigenetic regulators, such as miRNA and pluripotency factors, can be tightly integrated into molecular pathw...

Up-Regulation of Hepatitis C Virus Replication and Production by Inhibition of MEK/ERK Signaling

BACKGROUND Viruses interact with and exploit the host cellular machinery for their multiplication and propagation. The MEK/ERK signaling pathway positively regulates replication of many RNA viruses. However, whether and how this signaling pathway affects hepatitis C virus (HCV) replication and production is not well understood. METHODS AND RESULTS In this study, we took advantage of two well-...

Different Expression of Extracellular Signal-Regulated Kinases (ERK) 1/2 and Phospho-Erk Proteins in MBA-MB-231 and MCF-7 Cells after Chemotherapy with Doxorubicin or Docetaxel

Objective(s) Curative treatment of breast cancer patients using chemotherapy often fails as a result of intrinsic or acquired resistance of the tumor to the drug. ERK is one of the main components of the Ras/Raf/MEK/ERK cascade, which mediates signal from cell surface receptors to transcription factors to regulate different gene expression. In this study, cytotoxicity and the expression of Erk...

Gene Expression Profile Analysis during Mouse Tooth Development

Introduction: Complex molecular pathways involve in development of different tissues such as teeth. Differential gene expression patterns during teeth development generates different tooth types. Teeth development results from interactions between oral epithelium and underlying ectomesenchyme cells with neural crest origin. Teeth development are regulated by different signaling networks. In thi...
